FPbase was made by Talley Lambert from Harvard University as a one-stop shop for researchers to plan their imaging experiments.
The website has a trove of information on fluorescent proteins and dyes including sequences, spectra, evolution, lifetime, quantum yield etc.

Consider the following set up with mCherry + eGFP. Depending on your experience level and knowledge of how the microscopes work, you can add in light-sources, filters, and detectors to determine efficiencies as well.
